GWU STATISTICS SEMINAR SERIES

Title: Innovated Power Enhancement Tests for Complex and High Dimensional Data
Speaker: Lingzhou Xue, Penn State

Location: Duques Hall, Room 152 (2201 G St. NW, Washington, DC 20052)
Date & Time: April 20, Friday (11 AM-noon)

ABSTRACT: In the current literature, two sets of test statistics are commonly used for high-dimensional tests: 1) using extreme-value form statistics to test against sparse alternatives, and 2) using quadratic form statistics to test against dense alternatives. However, quadratic form statistics suffer from low power against sparse alternatives, and extreme-value form statistics suffer from low power against dense alternatives with small disturbances and may have size distortions due to its slow convergence. For real-world applications, it is important to derive powerful testing procedures against more general alternatives. Based on their joint limiting laws, we introduce a new general power enhancement testing procedure to boost the power against more general alternatives and retain the correct asymptotic size. Under the high-dimensional setting, we derive the closed-form limiting null distributions, and obtain their explicit rates of uniform convergence. The proposed power enhancement test is demonstrated in various parametric and nonparametric tests for high-dimensional means, banded covariances, spiked covariances, and so on. We demonstrate the finite-sample performance of our proposed testing procedures in both simulation studies and real applications.

GWU STATISTICS SEMINAR SERIES

TITLE: Semiparametric regression analysis for composite endpoints subject to component-wise censoring
SPEAKER: Guoqing Diao, George Mason University, Statistics

Location: DUQUES HALL, Room 254 (2201 G St. NW, Washington, DC 20052)
(please note the room change)

Date & Time: April 13, Friday (11 AM-noon)

ABSTRACT:

Composite endpoints with censored data are commonly used as study outcomes in clinical trials. For example, progression-free survival is a widely used composite endpoint, with disease progression and death as the two components. Progression-free survival time is often defined as the time from randomization to the earlier occurrence of disease progression or death from any cause. The censoring times of the two components could be different for patients not experiencing the endpoint event. Conventional approaches, such as taking the minimum of the censoring times of the two components as the censoring time for progression free survival time, may suffer from efficiency loss and could produce biased estimates of the treatment effect. We propose a new likelihood-based approach that decomposes the endpoints and models both the progression-free survival time and time from disease progression to death. The censoring times for different components are distinguished. The approach makes full use of available information and provides a direct and improved estimate of the treatment effect on progression-free survival time. Simulations demonstrate that the proposed method outperforms several other approaches and is robust to various model misspecifications. An application to a prostate cancer clinical trial is provided.
