Merck and Moderna have reported positive Phase III results for an investigational personalized mRNA therapy combined with immunotherapy for patients with melanoma at high risk of returning after surgery.

The findings build on earlier research, including a Phase II clinical trial in which Georgetown Lombardi was among the participating cancer centers.

“I was very excited to hear that the clinical trial results continue to show promise,” said Angela Evatt, who participated in the earlier trial through Georgetown Lombardi and MedStar Health. “With each release of the study results, it continues to give me hope that there is less of a likelihood of recurrence for me and others.”

New results from the I-SPY2 trial, published in JAMA Oncology, point to real promise in treating high-risk, early-stage HER2-negative breast cancer.

Georgetown Lombardi researchers, led by Dr. Claudine Isaacs, tested a combination of two immunotherapy drugs (cemiplimab and fianlimab) added to standard chemotherapy before surgery. The results are encouraging. Pathologic complete response rates roughly doubled, climbing from 21% with standard care to 44% with the new regimen for all HER2-negative patients. In triple-negative breast cancer, rates rose from 29% to 53%.

The combination carried added side effects; this specific regimen will not advance for further study. But the findings establish an important proof of principle for dual immune targeting, opening the door to newer drugs that could deliver similar benefit with fewer side effects.

As Dr. Isaacs says, “The bottom line is that we want to individually treat people with the drugs that they need and avoid toxicities for optimal outcomes.” Read more:
