【IMB研究成果發表-陳律佑副研究員】
癌細胞永生的秘密:孤兒核受體透過TRIM28 啟動端粒延長機制

一般細胞在分裂多次後，染色體端粒會逐漸縮短，最後細胞會停止分裂。然而，癌細胞會利用「替代性端粒延長機制(Alternative Lengthening of Telomeres, 簡稱ALT)維持端粒長度，避免細胞老化並持續分裂。

本院分生所陳律佑副研究員研究團隊發現，ALT的啟動與端粒上的「H3K9me3異質染色標記」密切相關，這種標記代表端粒DNA結構變得更為緊密，也被視為是ALT運作的關鍵。研究指出，孤兒核受體(orphan nuclear receptors)會將轉錄共抑制因子(TRIM28)帶到端粒位置，促使端粒染色質轉變為H3K9me3標記的異染色質狀態，進而改變端粒結構並啟動ALT機制，使癌細胞得以維持端粒長度且持續分裂。研究進一步發現，TRIM28是ALT活化的必要因子。除了孤兒核受體外，KRAB domain亦可透過TRIM28誘導類似ALT現象，顯示ALT啟動與染色質異質狀態密切相關。

論文第一作者為本院國際研究生學程「分子與細胞生物學」(TIGP-MCB)博士生蔡佳岑。研究經費由本院關鍵突破計畫與國科會共同支持。研究成果已於2026年3月31日發表於《歐洲分子生物學組織期刊》(The EMBO Journal)。
