A high-fat diet (HFD) leads to insulin resistance and impaired endothelial function, accompanied by dysfunction of the perivascular adipose tissue (PVAT). The mechanisms responsible for the reversal of endothelial dysfunction following a return to a standard diet remain poorly understood. Therefore, in this study, we examined the early and late effects of HFD withdrawal in a mouse model. After just one week of HFD withdrawal, we observed a reversal of systemic insulin resistance, but it was not accompanied by a complete recovery of endothelial function in the aorta, despite changes in the biochemical phenotype of PVAT with respect to lipid unsaturation. Complete reversal of endothelial dysfunction was only evident after 6 weeks of HFD withdrawal and was associated with changes in the guanylate cyclase pathway in PVAT. Overall, our study showed that the restoration of endothelial function in the aorta after HFD withdrawal is independent of the mechanisms of systemic insulin resistance and lipid unsaturation in PVAT and may be dependent on the guanylate cyclase pathway in PVAT, indicating potential new therapeutic options for restoring endothelial function in metabolic diseases.

W dniach 14-15 maja 2026 w Łodzi odbyła się konferencja BioOpen, podczas której nasza doktorantka Magda Machnik prezentowała poster pt. "Lipid composition changes in perivascular adipose tissue upon high-fat diet induced obesity: The role of stearoyl-Coenzyme A desaturase 1". Prezentacja spotkała się z uznaniem Komitetu Naukowego, który przyznał nagrodę za najlepszy poster w kategorii "Molecular and Medical Biology". Wyróżnione wyniki zostały uzyskane w ramach realizacji projektu OPUS27 "Heterogeneity of perivascular adipose tissue in the browning process: at the crossroads of single-cell Raman spectroscopy and pathophysiology of adipocytes" kierowanego przez dr Krzysztofa Czamarę.
