pH meter is used for measuring hydrogen ions concentration in water based solutions to check about acidic or alkaline behavior of solutions.
How to measure pH?
1) Rinse the electrode tip in deionized water to pH of glass membrane neutral
2) Enter the sensitive glass membrane of pH meter into the solution for which pH is to be measured.
3) Wait until the readings appear on meter

All broth ingredients are added when making LB Agar media, but the difference is that the pH is maintained before adding the agar. Agar (being a solidifying agent) gets solidified earlier due to which the pH cannot be normalized so the pH is always maintained first and then agar is added according to its requirements and autoclaved. Then it is poured on Petri plate for culturing.

LB Media (Broth)
LB, an abbreviation of Luria-Bertani, media is used for the culturing of bacteria, especially Escherichia Coli. It contains three major components.
For the preparation of 100ml media, we use
1) Tryptone 1g
2) Yeast Extract 0.5g
3) NaCl 1g
Apart from these components, a Master Mix, which contains all components in one recipe, can be used.
