Difference between the gram positive and grams negative bacteria is the presence of peptidoglycan layer which attracts stains during microscopy.
The primary stain which are attracted by the Gram positive bacteria are the crystal violet, carbon fuchsin, malachite green and methylene blue. While the counter stain which is being attracted by the Gram negative bacteria are safrarin and methylene blue.
