microbiology is the branch of biology that deal with study of microscopic organisms
microscopic organisms or microbes are organisms which can not be seen with our naked eyes except with the aid of microscope
five group of microorganisms
1- virus
2- bacteria
3- protozoa
4- fungi
5- algae
They are found every where in nature,
in soil, surface of an object, and within living organisms where they are able to invade a living cells
Culture: is the growing of microorganisms in a special medium in laboratory.
In other to grow a microorganisms we have to use a procedure that will not introduce microbial growth from our surroundings, such as air, clothes, apparatus, water, etc.
Bacteria, fungi, algae are easy to grow in a petri dish or test tube.
Even living tissue and cells of multicellular organisms can be culture for study in appropriate media- a technique know as tissue culture.
unfortunately virus can only be grow and multiply inside living organisms so they can not be grow like other microorganisms.
many disease cause organisms are difficult to grow
important of tissue culture
1- hereditary mechanisms
2- growth and development
3- details of cells structure and metabolism
4- nature of specific inherited disorders
use of tissue culture
1- for production of vaccine against viral disease
2- to diagnosis inherited disorders
3- for the culture of virus
Aseptic technique
these include:
1-sterilize all glass that will contact with specimen
2-wipe the work area with clothes or cotton wool socked in a strong disinfectant
3- flaming all equipments using for transfer of microorganisms just before used.
How to identify a microorganisms.
i- observing the form and structure of microorganisms
ii- characteristics of the colonies form
iii- oxygen requirements of microorganisms especially bacteria
those that requires oxygen are called aerobic bacteria and those that can grow in absence of oxygen are called obligated anaerobic bacteria
