1. Protein Separation: Proteins from the sample are separated based on their size using gel electrophoresis, typically SDS-PAGE.
2. Protein Transfer: The separated proteins are transferred from the gel to a solid membrane, such as nitrocellulose or PVDF.
3. Blocking: The membrane is treated with a blocking agent to prevent non-specific binding and reduce background noise.
4. Primary Antibody Incubation: The membrane is incubated with a primary antibody that recognizes the target protein.
5. Washing: The membrane is washed to remove unbound or non-specifically bound antibodies.
6. Secondary Antibody Incubation: The membrane is incubated with a secondary antibody that binds to the primary antibody.
7. Washing: The membrane is washed again to remove unbound secondary antibodies.
8. Detection: The presence of the target protein is visualized using a detectable marker conjugated to the secondary antibody. This can be done through chemiluminescence or fluorescence.
9. Analysis: The Western blot image is captured and analyzed to determine the abundance of the target protein.
