Proud Moment for Government Pharmacy College Sajong (GPCS), Sikkim (under the aegis of H&FW, Government of Sikkim, India).
Congratulations to Dr. Ankur Gupta, Associate Professor, Department of Pharmaceutical Chemistry, GPCS and his M.Pharm students, Ms. Pratiksha Khanal, and Ms. Anusha Chettri on their pioneering research on “Discovery of Piplartine as a Potent Multitargeting Anti-HIV and Anti-TB Lead Molecule: Network Pharmacology, In silico, and In vitro Studies to Analyse Its Role in HIV/TB Co-infection” published in Chemistry Europe’s, Wiley Journal, ChemistrySelect. The work was carried out in collaboration with Dr. Anupam Mukherjee from ICMR-National Institute of Virology, Dr. Nisheeth Agarwal, BRIC-THSTI, and scientists from ICMR-National Institute of Translational Virology and AIDS Research.
The Authors and GPCS would like to show their gratitude with a heartfelt thanks to the Health Secretary, the Principal Director, and the Director, Health & Family Welfare (H&FW) Department, Government of Sikkim, India for their endless support and encouragement for this work.
This study investigated the Piplartine’s therapeutic potential against HIV, Mtb, and co-infection using network pharmacology, in silico, and in vitro approaches. Gene Ontology and KEGG pathway analyses highlighted TP53, STAT3, HSP90AA1, IL1B, and CASP3 as key regulators. Further, PL was isolated which exhibited potent anti-HIV activity with an EC₅₀ of 0.735 μg/mL in TZM-bl cells. It inhibited HIV-1 replication in strains HIV-1VB028 and HIV-1UG070. PL inhibited RTase and Protease activity by 75% and 58.6%, respectively, and lowered p24 antigen levels by ~84% at day 5 post-infection. Viral load dropped to 66,180 copies/mL by day 11, compared to >10,00,000 copies/mL in controls. Additionally, PL showed antimicrobial activity against Mtb H37Rv with an MIC of 37.5 μg/mL, highlighting its potential as a promising lead candidate against HIV/Mtb co-infections.
