Important stuff….

People need to trip to have therapeutic outcomes. The science is clear; the quality of the acute psychedelic experience has been shown to predict therapeutic outcomes after psychedelic therapy.

Taking the trip out of psychedelics might make them easier to sell, prescribe, and market. But the trip is where much of the mind-revealing aspect of the psychedelic experience happens. It is where emotional release can surface, and where profound shifts in perspective may take place.

Remove the trip, and you may also remove a central part of what makes psychedelics therapeutically transformative.

Eli Lilly, maker of Zepbound and Mounjaro, is buying AtaiBeckley for up to $3.8 billion (roughly $2.8 billion upfront, plus $1 billion in milestones), marking one of the biggest bets yet on psychedelic medicine.

The deal hands Lilly BPL-003, a DMT-based nasal spray in late-stage trials for treatment-resistant depression, alongside another DMT compound and an MDMA-related asset, a full pipeline rather than a single drug.

Analysts see BPL-003 alone generating $1-2 billion in sales if trials succeed, with results expected in early 2029. The wider psychedelics market could hit $12 billion by 2034, rivaling today's branded antidepressant market (about $8 billion), per RBC Capital Markets.

The move follows Trump's April executive order speeding regulatory review of psychedelic treatments, and echoes AbbVie's 2025 deal for a Gilgamesh Pharmaceuticals compound. Shares of Enveric Biosciences and GH Research jumped on the news.

For decades, psychedelics were illegal, so there was no money in it for pharma. That left the underground, facilitators, researchers, and communities working outside the law, to carry the research, refine the practice, and prove the healing potential long before anyone in a boardroom cared.

So what do you make of this? Is it overall a positive that psychedelics are being commercialised and medicalised by big pharma? On one hand, it means wider access. On the other, it risks diluting traditional wisdom and overlooking the underground communities who risked their freedom to get us here.

World’s Largest Pharmaceutical Company, Eli Lilly, Acquires 5-MeO-DMT Developer AtaiBeckley for Up to $3.8 billion

This morning, Eli Lilly confirmed that it will acquire AtaiBeckley, following rumours circulated in Bloomberg yesterday, as well as our reporting last week that a deal was nearing completion.

AtaiBeckley’s lead candidate is intranasal 5-MeO-DMT (BPL-003) for treatment-resistant depression, which is now entering Phase 3. It has been competing fiercely with GH Research, which is developing an inhaled formulation of the drug for the same condition.

Lilly will pay $2.8 billion in cash upfront, with potential for an additional $1 billion in milestone-based biobucks over the coming years. That makes it the largest psychedelic buyout, by quite some margin, though some investors Psychedelic Alpha spoke with were disappointed by the terms.

Voices within the psychedelics field are mixed, too. Some have welcomed the acquisition as a sign of the field’s maturity, while others are skeptical of the pharmaceutical juggernaut’s fit for stewarding the development and potential roll-out of a potent psychedelic like 5-MeO-DMT.

The transaction is expected to close this quarter.

More research, more legitimacy, more funding. More access. And potentially, eventually, more treatments reaching people that could profoundly change their lives. But at what cost?

A phase 2a open-label trial published in the Journal of Clinical Psychiatry tested inhaled 5-MeO-DMT (GH001) in ten women with moderate-to-severe postpartum depression.

Using an individualised dosing regimen of up to three escalating doses (6mg, 12mg, 18mg) on a single day, researchers found a mean MADRS reduction of 35.4 points by day 8, corresponding to approximately 96% symptom reduction.

All ten patients achieved full remission, with effects observable just two hours post-dose. Maternal functioning improved by 56%. Suicidal ideation dropped from three patients to zero by discharge. No serious adverse events were reported and all patients were discharged the same day.

Breast milk analysis in lactating patients showed mebufotenin cleared within approximately eight hours. The compound acts as a nonselective serotonin agonist with preferential 5-HT1A affinity and a short plasma half-life, producing a psychoactive window of roughly 20 to 25 minutes per dose.

No psychotherapy was included. Limitations include small sample size, no control arm, and one-week follow-up.

A fascinating preprint about traditional psilocybin use outside the Americas:

“Our study followed an anecdotal report from a Basotho healer, collected by Mamosebetsi Sethathi and published in 2024, who claimed to use the mushroom Psilocybe maluti in a divination ritual. This potentially constituted the first case of traditional psilocybin use outside the Americas, but the initial evidence was weak.”

“Unlike the large ritualized doses characteristic of Mesoamerican psilocybin use, Basotho healers primarily apply small doses of P. maluti alongside other psychoactive plants, most notably the hallucinogenic bulb Boophone disticha. Multiple lines of evidence suggest these practices predate the mid-twentieth-century popularization of psilocybin. These findings expand the known geographic scope of traditional psilocybin use and reveal a mode of psychedelic application distinct from existing ethnographic models.”

Interviewees reported uses in four categories: initiation, healing, recreation, and magical protection.
