Groundbreaking medical research led by Dr. Mariano Barbacid at Spain’s National Cancer Research Centre (CNIO) has successfully achieved the complete and permanent elimination of pancreatic tumors in mouse models.
The study, published in January 2026 in the journal PNAS, utilized a innovative triple-drug combination therapy designed to bypass the tumor's typical resistance mechanisms.
This "triad" approach targets the KRAS oncogene pathway from three distinct angles: directly inhibiting the mutated KRAS protein, shutting down escape routes like the EGFR and HER2 pathways, and disabling the STAT3 stress-response system.
In experimental settings, this therapy led to full tumor regression with no recurrence for over 200 days post-treatment and notably low toxicity.
While these results mark a monumental step in oncology research, experts caution that the therapy has not yet entered human clinical trials and requires further validation for safety and efficacy in patients.
The breakthrough offers a new blueprint for treating pancreatic ductal adenocarcinoma, one of the world's most lethal and treatment-resistant cancers.
