So here's the follow-up to the stem cell project... my biotech research students cultured the MSCs to confluency (>80% plus proliferation in the flask). They then determined an estimated number of cells using a hemocytometry and microscope. We finished by passaging the stem cells (determining the appropriate concentration of cells and dilution factor for the media going into new flasks) to keep the cell line going. Next step - metabolic assays!
